Mechanical Strain and Microtubule-associated Signaling in the Lung Endothelium

چکیده
This study demonstrates that pathological cyclic stretch (CS) alter microtubule (MT) stability and induces MT reorientation. MT stabilization inhibits stress fiber formation and delays cell alignment in response to pathological CS. MT-associated GEF-H1 knockdown attenuates stretch-induced rearrangement of actin skeleton and controls stretch-dependent MT dynamics in pulmonary endothelium (EC). GEF-H1 depletion and MT stabilization attenuate ventilator-induced lung injury in vivo.

Mechanotransduction by GEF-H1 as a novel mechanism of ventilator-induced vascular endothelial permeability.
Pathological lung overdistention associated with mechanical ventilation at high tidal volumes (ventilator-induced lung injury; VILI) compromises endothelial cell (EC) barrier leading to development of pulmonary edema and increased morbidity and mortality. We have previously shown involvement of microtubule (MT)-associated Rho-specific guanine nucleotide exchange factor GEF-H1 in the agonist-ind...

Microtubules are linear filaments of the cytoskeleton that mediate such cellular processes as nerve growth and cell division and in general serve to organize the cytoplasm. In interphase cells microtubules tend to exhibit a radial pattern with the plus ends of microtubules pointing outward toward the cell periphery and the minus ends pointing inward toward the centrosome, near the nucleus. Dist...

Transmission of externally applied mechanical forces to the interior of a cell requires coordination of biochemical signaling pathways with changes in cytoskeletal assembly and organization. In this study, we addressed one potential mechanism for this signal integration by applying uniform single external mechanical strains to aortic smooth muscle cells (SMCs) via their adhesion substrate. A te...
متن کاملFree Vibration Analysis of Microtubules as Orthotropic Elastic Shells Using Stress and Strain Gradient Elasticity Theory
In this paper, vibration of the protein microtubule, one of the most important intracellular elements serving as one of the common components among nanotechnology, biotechnology and mechanics, is investigated using stress and strain gradient elasticity theory and orthotropic elastic shells model. Microtubules in the cell are influenced by internal and external stimulation and play a part in con...

Microtubules, the active filaments with tubular shapes, play important roles in a wide range of cellular functions, including structural supports, mitosis, cytokinesis, and vesicular transport, which are essential for the growth and division of eukaryotic cells. Finding properties of microtubules is one of the main concerns of scientists. This work helps to obtain mechanical properties of m...
